Naproxen and esomeprazole magnesium

Mechanism of action: NSAID + proton pump inhibitor (fixed-dose combination)
In plain language: Combines a pain-and-inflammation reliever with a stomach-acid blocker in one tablet: naproxen, an anti-inflammatory that can irritate the stomach lining, and esomeprazole, which suppresses stomach acid to protect against that irritation.
How it works: VIMOVO combines an enteric-coated naproxen core with an immediate-release esomeprazole magnesium layer, designed so esomeprazole is released first in the stomach, ahead of naproxen, which doesn't dissolve until it reaches the small intestine. Per the FDA label, naproxen is an NSAID whose analgesic and antipyretic effects are believed to work through inhibition of prostaglandin synthesis; esomeprazole is a proton pump inhibitor that suppresses gastric acid secretion by specifically inhibiting the H+/K+-ATPase in the parietal cell. The combination doesn't change naproxen's anti-inflammatory action -- it pairs it with acid suppression specifically to lower the risk of NSAID-associated gastric ulcers.
Therapeutic applications: VIMOVO is FDA-indicated for relief of the signs and symptoms of osteoarthritis, rheumatoid arthritis, and ankylosing spondylitis, and to decrease the risk of developing gastric ulcers in patients at risk of developing NSAID-associated gastric ulcers.
